I want to add a chamber fan to my gun, and seal the back plate on, but, doest the back plate have to be off when inserting a projectile, so the air doesn't compress in the chamber?

what back plate? a plug? that should screw on and off. i think it was you who just made a nice propane gun. you still need to unscrew the back cap to vent it after each shot.

If he has a chamber fan with propane as fuel then he can vent it through the barrel, right?

no, not really. where would it vent to? exactly, it would just move around in the chamber and not go anywhere. after maybe 3 or 4 minutes it would, but if the plug came off it would vent in a few seconds.
